Background :
Dipeptidyl peptidase 3 (DPP3) is a zinc-dependent peptidase distributed within the cytoplasm which cleaves dipeptides sequentially from the N-terminus of bioactive peptide substrates 4+ amino acid residues in length. DPP3 adopts a bi-lobed structure and features HEXXGH and EEXRAE/D motifs which coordinate the Zn2+ ion in its active site. DPP3’s gene is located on chromosome 11q12-13.113. DPP3 regulates the transformation of enkephalin, opioid peptide, terminal protein, and cell signaling. Deficiency results in ROS formation in the kidneys via elevated Angiotensin II (DPP3’s most prominent reported substrate), III, IV, and ANG (1-5). DPP3 is linked to the development of cardiovascular diseases through RAS, oxidative stress, and inflammation pathways. Depletion was associated with the upregulation of proapoptotic proteins SMAC and p53, making it a potential target in tumor therapy.
