Background :
C-type Lectin Domain Family 4 Member C (CLEC4C) is a transmembrane protein selectively expressed on plasmocytoid dendritic cells (pDCs) which regulates secretion of proinflammatory cytokines and type I interferons in pDCs exposed to TLR7 and 9 agonists such as microbial CpG DNA. Its gene located on chromosome 12p13.31, CLEC4C consists of a 21 amino acid (aa) N- cytoplasmic domain, a 23 aa transmembrane segment, and a 169 aa extracellular domain that contains a juxtamembrane neck region and a C-carbohydrate recognition domain. Blastic plasmacytoid dendritic cell neoplasm is positive specifically for CLEC4C. Monoclonal antibodies directed against CLEC4C have achieved disease control in patients with cutaneous lupus erythematosus. CLEC4C-mediated suppression of IFNα production is regulated by the masking/unmasking of galactose moieties, representing a new potential strategy for regulating immune responses in pathological conditions.
